            湯亞杰,男,1973年10月出生,湖北武穴人,中共黨員,教授,博士生導(dǎo)師。2003年于華東理工大學(xué)生物反應(yīng)器工程國家重點(diǎn)實(shí)驗(yàn)室獲生物化工博士學(xué)位,2005年從美國麻省理工學(xué)院(MIT)學(xué)成回國,長期從事“天然產(chǎn)物生物智造”領(lǐng)域的教學(xué)科研工作。[1]

            湯亞杰教授主持了科技部“中青年科技創(chuàng)新領(lǐng)才”、教育部“新世紀(jì)優(yōu)秀人才支持計(jì)劃”、湖北省“楚天學(xué)者”獎勵計(jì)劃、國家自然科學(xué)基金、湖北省高端人才引領(lǐng)培養(yǎng)計(jì)劃等30多項(xiàng)科研課題。以通訊或第一作者在發(fā)酵工程領(lǐng)域主流學(xué)術(shù)期刊(如Scientific Reports、Metab Eng、Appl Environ Microbiol)上發(fā)表SCI論文58篇;單篇SCI他引最高達(dá)93次。以第一發(fā)明人授權(quán)發(fā)明專利18項(xiàng),含美國專利2項(xiàng)。獲湖北省科技進(jìn)步一等獎1項(xiàng)、教育部自然科學(xué)一等獎2項(xiàng)、湖北省自然科學(xué)二等獎1項(xiàng)、上海市自然科學(xué)二等獎1項(xiàng)。

            本著“高標(biāo)準(zhǔn)、嚴(yán)要求;送出去、請進(jìn)來”的原則,湯亞杰時刻以自己的一言一行影響著團(tuán)隊(duì)成員和學(xué)生,幫助他們養(yǎng)成優(yōu)良學(xué)風(fēng),形成獨(dú)立思考、獨(dú)立工作的良好習(xí)慣。堅(jiān)持科研反哺教學(xué)和“以人才培養(yǎng)為中心,在科學(xué)研究過程中培養(yǎng)創(chuàng)新型人才”的教學(xué)思想和理念。湯亞杰把“人的發(fā)展”作為本科生教學(xué)、研究生培養(yǎng)和青年教師扶持的根本目標(biāo)。[2]正是由于這種創(chuàng)新型研究生培養(yǎng)模式的成功,不僅使實(shí)驗(yàn)室取得豐碩的科研成果,更重要的是逐漸打造了一個創(chuàng)新團(tuán)隊(duì),武漢大學(xué)、華中科技大學(xué)、山東大學(xué)、武漢理工大學(xué)、華中農(nóng)業(yè)大學(xué)、陜西師范大學(xué)等985、211院校的本科生慕名來攻讀碩士學(xué)位,提升了湖北工業(yè)大學(xué)人才培養(yǎng)的質(zhì)量與品牌。

            湯亞杰應(yīng)邀出任“Bioprocess and Biosystems Engineering (Springer)”副主編(Associate Editor)、Biotechnology Journal (Wiley)的Advisory Board (顧問委員會)、BioMed Rearch International?(Hindawi)的Editorial Board (編輯委員會)。

            科研成果

            湯亞杰長期從事“發(fā)酵工程”領(lǐng)域的科學(xué)研究,他所帶領(lǐng)的學(xué)術(shù)團(tuán)隊(duì)以國家戰(zhàn)略需求產(chǎn)品為導(dǎo)向,將發(fā)酵工程三大關(guān)鍵科學(xué)問題,即:微生物菌種篩選和遺傳改造(上游)、發(fā)酵過程優(yōu)化與放大(中游)、發(fā)酵產(chǎn)物分析檢測與分離純化(下游),串聯(lián)起來,聯(lián)合上游、中游、下游三個方向研究力量,致力于解決實(shí)驗(yàn)室的生物技術(shù)成果產(chǎn)業(yè)化的瓶頸問題。

            (1)?黑孢塊菌液體深層發(fā)酵:創(chuàng)建了珍貴菌物資源——黑孢塊菌液體深層發(fā)酵高效生產(chǎn)菌絲體及生物活性物質(zhì)的過程,使塊菌生產(chǎn)周期由野生的4-12年縮短為發(fā)酵的5-10天,并闡明了發(fā)酵塊菌在香味、鮮味和營養(yǎng)組分相似或優(yōu)越于野生塊菌,證明了發(fā)酵塊菌可替代野生塊菌,為解決塊菌資源短缺提供了新思路。上述研究獲得國家自然科學(xué)基金(21376066、21206035)、教育部新世紀(jì)優(yōu)秀人才支持計(jì)劃、湖北省“楚天學(xué)者”計(jì)劃等資助;部分成果發(fā)表在發(fā)酵工程領(lǐng)域國際知名學(xué)術(shù)期刊雜志上,如Bioresource Technology?(IF=5.039)、Analytica Chimica Acta?(IF=4.517)、Applied Microbiology and Biotechnology(IF=3.811),獲湖北省自然科學(xué)二等獎1項(xiàng),授權(quán)發(fā)明專利4項(xiàng)。

            (2)?鬼臼類抗腫瘤天然藥物研究:構(gòu)建了定向修飾鬼臼類化合物分子結(jié)構(gòu)開發(fā)抗腫瘤新藥的技術(shù)平臺,建立了結(jié)構(gòu)新穎和分子多樣的“鬼臼類化合物庫”,并從中篩選出一系列具有成藥前景的新化合物。其中,與已應(yīng)用于臨床的鬼臼類抗腫瘤藥物“依托泊苷”相比,4-MP-PTOX和4-TG-PTOX對人胃癌細(xì)胞株的抑制活性分別提高了121和91倍,對人正常腎小管細(xì)胞的細(xì)胞毒性分別降低了19和17倍,水溶性分別提高了300%和124%;同時,4-MP-PTOX和4-TG-PTOX具有較強(qiáng)的微管蛋白靶向結(jié)合能力,與靶蛋白微管蛋白的結(jié)合常數(shù)分別為8.7和8.5 μM,較“依托泊苷”(15.9 μM)均提高了近100%,有望開發(fā)為新型微管蛋白聚合抑制性抗腫瘤藥物。上述研究獲得科技部中青年科技創(chuàng)新領(lǐng)才、國家自然科學(xué)基金(20976038、20706012)等資助;部分成果發(fā)表在Bioresource Technology?(IF=5.039)、Applied and Environmental Microbiology?(IF=3.952)、Current Medicinal Chemistry(IF=4.070) 等國際知名學(xué)術(shù)期刊上;授權(quán)發(fā)明專利6項(xiàng),含美國專利1項(xiàng)。

            (3)?琥珀酸生物制造:建立了微生物菌種基因工程分子操作平臺,對產(chǎn)琥珀酸大腸桿菌基因工程菌的代謝支路中關(guān)鍵酶基因進(jìn)行了定向改造,減少了副產(chǎn)物合成及磷酸烯醇式丙酮酸的代謝流損失,增強(qiáng)了琥珀酸生物合成,琥珀酸產(chǎn)量提高了5倍;同時解析了琥珀酸生物合成代謝網(wǎng)絡(luò),篩選出影響琥珀酸通量大小的關(guān)鍵節(jié)點(diǎn),為高產(chǎn)琥珀酸大腸桿菌的改造提供了新思路。上述研究獲得國家自然科學(xué)基金(21176059)、湖北省自然科學(xué)基金創(chuàng)新群體等資助,部分成果發(fā)表在Metabolic Engineering(IF=8.258)、Microbial Cell Factories?(IF=4.250)等國際知名學(xué)術(shù)期刊上。
